The second of four children, Elizabeth Claire Kemper, who would later be popular as Ellie Kemper, was born on May 2, 1980, in Kansas City, Missouri, to Dorothy Ann (Jannarone) and David Woods Kemper. At age 19, she was named the Queen of St. Louis's annual Veiled Prophet Ball. In 2002, Ellie graduated from Princeton University with a degree in English. She then spent a year at Oxford University studying English.

On July 7, 2012, Ellie married fiance Michael Koman, a former writer on “Late Night with Conan O'Brien” and the co-creator of the Adult Swim series “Eagleheart” and Comedy Central series “Nathan for You.” Ellie is the older sister of television writer Carrie Kemper. She is of Italian (from her maternal grandfather), English, Scottish and German lineage.
